Peyer's patches or aggregated lymphoid nodules are organized lymphoid follicles , named after the 17th-century Swiss anatomist Johann Conrad Peyer. They are an important part of gut associated lymphoid tissue usually found in humans in the lowest portion of the small intestine , mainly in the distal jejunum and the ileum , but also could be detected in the duodenum. Peyer's patches are observable as elongated thickenings of the intestinal epithelium measuring a few centimeters in length.

The functional maturation of M cells is dramatically reduced in the Peyer’s patches of aged mice

Healthy and adequate number of M cells is important for an effective intestinal barrier. Reduced Claudin 4 is responsible for increased paracellular transport of antigenic materials. Calprotectin is an inflammatory marker secreted by neutrophils during inflammation. Its level is considered specific for intestinal inflammation. After 6 weeks of consuming the respective diets, stool and GIT segments containing PP were collected.

After tissue processing, tissues were sectioned into 3 micrometer thickness and were taken on poly-L-lysine coated glass slides. Immunohistochemical staining was done by rat M cell specific CK-8 antibody and anti-Claudin 4 antibody. Scoring was done to calculate the average number of M cells and Claudin 4 in the PP of both groups under light microscope.

Statistical analysis was done by chi-square test and independent T-test. High dietary fat selectively increases catalase expression within cardiac mitochondria. J Biol Chem. Ge, J. Why does a high-fat diet induce preeclampsia-like symptoms in pregnant rats.

Peyer's patch

Microfold M cells are located in the epithelium covering mucosa-associated lymphoid tissues, such as the Peyer's patches PPs of the small intestine. M cells actively transport luminal antigens to the underlying lymphoid follicles to initiate an immune response. The molecular machinery of M-cell differentiation and function has been vigorously investigated over the last decade. Studies have shed light on the role of M cells in the mucosal immune system and have revealed that antigen uptake by M cells contributes to not only mucosal but also systemic immune responses. However, M-cell studies usually focus on infectious diseases; the contribution of M cells to autoimmune diseases has remained largely unexplored.

The lumen of gastrointestinal tract is exposed to several potentially pathogenic microorganisms, thus it is extremely relevant to understand how immunosurveilance can be established. PPs contain a high percentage of B and T lymphocytes, macrophages and dendritic cells. Here we summarize a protocol for isolation and culture of mouse PP cells, which can be used to get a better insight into immunopathologies of microbes and to evaluate immune responses elicited by mucosal vaccines. The microfold M cell residing in the follicle-associated epithelium is a specialized epithelial cell that initiates mucosal immune responses by sampling luminal antigens. The differentiation process of M cells remains unclear due to limitations of analytical methods. Here we found that M cells were classified into two functionally different subtypes based on the expression of Glycoprotein 2 GP2 by newly developed image cytometric analysis.

By their ability to transport luminal antigens and bacteria, PPs can be considered as the immune sensors of the intestine. PPs functions like induction of immune tolerance or defense against pathogens result from the complex interplay between immune cells located in the lymphoid follicles and the follicle-associated epithelium. This crosstalk seems to be regulated by pathogen recognition receptors, especially Nod2. In turn, CD T-cells present in the PP are able to modulate the paracellular and transcellular permeabilities.

Microfold cells or M cells are found in the gut-associated lymphoid tissue GALT of the Peyer's patches in the small intestine , and in the mucosa-associated lymphoid tissue MALT of other parts of the gastrointestinal tract.

The mouse caecal patch is located near the blind end of the caecum, and consists of a group of lymphoid follicles.
